On the cover: In this ink-wash landscape, ten blazing suns hang overhead, scorching the land and withering living forms, as discussed by Zheng et al. in this issue of Neuron, which shows that sustained activation of the mitochondrial integrated stress response (mtISR) drives neuromuscular pathology in ALS. Layered cloud textures resemble mitochondrial cristae, suggesting a mitochondrial origin of disease. The multiple suns symbolize elevated PGAM5 protein levels, and their collective intensity reflects chronic mtISR activation. Under sustained exposure, sun-facing branches wither and leaves fall, mirroring neuromuscular junction deterioration. The scene conveys a loss of balance, in which a stress pathway intended to preserve cellular homeostasis becomes persistently engaged and ultimately accelerates neuromuscular system decline.
